Elucidating the Role of ROBO1 in NOTCH Activation Within the Mammary Gland

This research was conducted to better understand the role of ROBO1 in Notch activation during mammary gland development. The environment of the luminal compartment and the basal compartment of the mammary gland was replicated in vitro by culturing HC11 and MDA-MB231 cells. I investigated ligand expression in both cell lines through Western blotting. In HC11 cells, there was little to no change in Notch ligand expression or upstream targets of the AKT pathway. In MDA-MD231s, there was an increase in JAGGED1 expression and a decrease in Delta-Like 1 expression in the Robo1 knock down. This suggests that Robo1 may be regulating Notch ligand levels in order to govern Notch activation and ultimately alveologenesis.
